| Function | Tumor necrosis factor receptor superfamily member 4 (TNFRSF4), also known as OX40; a T-cell secondary costimulatory molecule which enhances proliferation, cytokine production and survival; its ligand OX40L, or CD252, binds to OX40 receptors on T-cells, preventing them from dying and subsequently increasing cytokine production. Without CD28, OX40 expression is delayed and reduced. OX40 also binds TRAF2, 3 and 5 and PI3K. OX40 gene mutations can cause a type of immunodeficiency. Roche has an experimental monoclonal antibody in development that targets OX40 (RG7888); anti-OX40 functions as an agonist antibody, which results in activation rather than blockade of the OX40 signaling pathway upon receptor binding. |
