Overview of Organoid Culture Media

Organoid culture requires a combination of basal medium with cytokines, amino acids, small molecule inhibitors, and other substances. The conditions vary based on tissue source. Many organoids are cultured in Matrigel, a mouse sarcoma-derived ECM-rich material, supplemented with growth factors. While Matrigel is versatile and economical, its complex composition (over 1800 proteins) complicates identification of signals needed for proper organoid formation. Moreover, it may lack essential components for certain organoids, and its mouse origin poses immunogenic risks for human clinical applications.

Organoid Medium Growth Factor Formulation

A key component of organoid media is the availability of essential growth factors. Typically, the production of growth factors relies on eukaryotic expression systems to ensure proper disulfide bond formation and macromolecular folding. In the eukaryotic expression system, growth factors are secreted from the expression host into the cell culture medium. This conditioned medium containing the selected growth factors is then diluted directly into organoid medium for application. The growth factors required for different organoid cultures are not exactly the same. For example, intestinal organoids require the addition of EGF, Noggin, R-Spondin-1, and Wnt-3a basic growth factors, whereas brain organoids require FGF- basic, Noggin, DKK-1, EGF, BDNF, and GDNF basic growth factors.

Development of Organoid Culture Media

With the development of organoid culture technology, organoid culture media are also being innovated. Currently, the development of novel matrices such as decellularized extracellular matrix, synthetic hydrogels, gel-forming recombinant proteins, etc. provides new ideas for organoid culture solutions. Among them, decellularized methods have been proven to be suitable for the culture of human kidney, mouse kidney, mouse heart, human and porcine lung, and porcine testicular tissue. Synthetic hydrogels and gel-forming recombinant proteins have been characterized by mechanical properties, controllability of function and erosion rates, and programmability of MMP degradation enzyme recognition sites. Recombinant ECM proteins have been investigated for use in pancreatic-like organ cultures.
