Overview of Skin Tissue Engineering

An important means of repairing skin defects is autologous thick layer implants, but this method has some obvious limitations, such as new damage to the skin donor area and insufficient skin sources. Therefore, the development of tissue-engineered skin for permanent replacement of skin defects is a desirable goal. After decades of development, a variety of tissue-engineered skins have been put on the market for application, but there is still a long way to go to achieve the regenerative repair of skin defects.

  • Three Main Elements for Skin Tissue Engineering
    The three main elements for building tissue-engineered skin are seed cells, growth factors, and scaffold materials. Tissue-engineered skin can be formed by expanding a large number of functional cells in vitro in culture and then compounding them onto a three-dimensional (3D) polymeric scaffold material. The scaffold contains functional growth factors. The cells interact with the scaffold to induce and grow to form a three-dimensional, active skin substitute.

Advances in Skin Regeneration Using Tissue Engineering

Artificial skin is by far the most successful tissue-engineered material in clinical applications and is the first commercially available product in tissue engineering. However, tissue-engineered skin is still a long way from being a truly ideal permanent skin substitute, as it is not yet possible to construct skin appendage structures such as hair follicles and sweat glands. Currently, the main role of artificial skin is to close skin wounds, accelerate healing and stimulate autologous skin tissue regeneration and epithelialization for therapeutic purposes.

Applications of Tissue-engineered Skin

Tissue-engineered skin is generally used for wound healing. Its indications mainly include acute large skin defects, such as large burns and scalds, and various types of chronic wounds of the skin, such as chronic refractory ulcers after trauma, venous ulcers of the lower extremities, and diabetic extremity ulcers. In addition, tissue-engineered skin can also be used for in vitro testing of cosmetics, chemicals, drugs, medical devices, and textiles
