Organoids as Improved Model for Disease Research

Organoids more accurately model human gut function and physiology than animal models or 2D models. Organoids are models with a three-dimensional structure consisting of pluripotent tissue-specific stem cells and their more differentiated progeny.

Currently, organoids have been shown to be widely used in experimental settings. Because they are compatible with routinely used imaging techniques as well as live cell imaging, and are capable of metabolic activity assessment, genome-wide expression, protein analysis, gene editing, and high-throughput assays. As a result, organoids are increasingly used to study organ tissue function, model disease phenotypes, and drug discovery screens.

Elements for Organoids Culture

Organoids are derived from pluripotent stem cells or adult stem cells, also known as tissue stem cells. Typically, the process of organoid formation involves several key points. First, activation or inhibition of key signaling pathways that regulate developmental patterns to properly establish the direction of stem cell differentiation. Secondly, the development of a medium containing growth factors and nutrients to meet the normal growth and differentiation of all cell types within the organoid. Finally, to provide three-dimensional growth space for cell aggregation by embedding three-dimensional scaffolds.
