Culture medium for long-term expansion and maintenance of human liver organoids

| Overview | Reliably expand, and maintain human liver organoids long-term as a model system for studying liver biology, disease modeling, drug discovery, and therapeutic screening. Use Organoid Growth Medium (OGM) (Human) to establish hepatic organoids from human liver tissue, or PSC-derived hepatic cells generated with the Hepatocyte Kit. Available either individually or as part of Organoid Kit (Human), OGM can be used to culture and expand liver organoids for future experimentation or bio-banking. Organoids initiated in Organoid Initiation Medium (Human) and expanded in OGM express self-renewal (LGR5, AXIN2), ductal (KRT19, SOX9, KRT7), and hepatic progenitor (HNF4A) markers. These organoids can also be differentiated using Organoid Differentiation Medium (Human) to mature organoids that demonstrate hepatic functionality, including CYP3A4 activity. Use OGM to culture organoids in a range of formats, including in domes for routine culture, in dilute suspensions in spinner flasks for scale-up, in 96-well plates for high-throughput screening applications, and as organoid-derived two-dimensional monolayers on inserts for disease-modeling applications. The organoids can also be cryopreserved for later use, remaining highly viable and retaining their capacity for further differentiation when thawed and cultured in OGM. |
